Poor invasion of trophoblastic cells but normal plaque formation in fibroblastic cells despite actA deletion in a group of Listeria monocytogenes strains persisting in some food processing environments.

چکیده
We determined mammalian cell invasion and virulence gene (inlA, inlB, and actA) sequences of Listeria monocytogenes strains belonging to a molecular subtype (RAPD 9) that often persists in Danish fish-processing plants. These strains invaded human placental trophoblasts less efficiently than other L. monocytogenes strains, including clinical strains, and they carry a premature stop codon in inlA. Eight of 15 strains, including the RAPD 9 and maternofetal strains, had a 105-nucleotide deletion in actA that did not affect cell-to-cell spread in mouse fibroblasts. The RAPD 9 strains may still be regarded as of low virulence with respect to human listeriosis.

ActA is required for crossing of the fetoplacental barrier by Listeria monocytogenes.
The facultative intracellular bacterial pathogen Listeria monocytogenes induces severe fetal infection during pregnancy. Little is known about the molecular mechanisms allowing the maternofetal transmission of bacteria. In this work, we studied fetoplacental invasion by infecting mice with various mutants lacking virulence factors involved in the intracellular life cycle of L. monocytogenes. We...
متن کاملInhibitory Effect of Nisin on Listeria monocytogenes Inoculated into Surimi and Minced Meat
Background & Objective: Listeria monocytogenes has already established as an important food born pathogen which induce listeriosis in human. Use of bacteriocins to provide food safety has been increased dramatically. Nisin has a wide spectrum inhibitory effect than the other bacteriocins and inhibits food-borne pathogens such as L. monocytogenes and many other Gram-positive spoilage microorg...
متن کاملBiofilm Formation and Antibacterial Properties of Lactobacillus Isolated from Indigenous Dairy Products
Background: The health benefits of probiotic bacteria are not unknown to anyone. On the other hand, indigenous dairy sources are a potential source of native probiotics. This study aimed to describe the inhibitory activity of Cell-Free Supernatant (CFS), planktonic cells, and biofilm form of Lactobacillus strains isolated from native dairy sources on food pathogens. Methods: Antibacterial acti...
متن کاملMolecular Identification of Listeria monocytogenes in Raw Hamburgers from Kerman, South-East of Iran
Background: Listeria monocytogenes is a Gram-positive and facultative anaerobic food-borne bacterium which is capable of intra and extra cellular growth. L. monocytogenes usually can exist on different surfaces and instruments at production and processing sites of food products with animal origin. In Iran, the consumption of burger has increased recently, but its safety is still of great concer...
متن کاملEffect of octenidine hydrochloride on planktonic cells and biofilms of Listeria monocytogenes.
Listeria monocytogenes is a food-borne pathogen capable of forming biofilms and persisting in food processing environments for extended periods of time, thereby potentially contaminating foods. The efficacy of octenidine hydrochloride (OH) for inactivating planktonic cells and preformed biofilms of L. monocytogenes was investigated at 37, 21, 8, and 4 degrees C in the presence and absence of or...
